For Defense, Yo-Yo, Quake, Assault, Medic, and Coulson is probably the best option. Yo-Yo loses her passive Offense Down if there's another Protector, which means no Fury security summons or either Captain America. Medic will still always revive Coulson, he'll still proc all the either SHIELD toons, and his bonuses are exclusively Defense oriented.
